小分子化合物N-414抑制胃癌细胞侵袭及机制研究

目的 研究小分子化合物N-414对人胃癌细胞SGC7901、BGC823增殖、侵袭能力的影响及其作用机制。方法 应用MTT检测不同浓度小分子化合物N-414对于胃癌细胞SGC7901和BGC823增殖能力的影响。分别用0、20、40、60μmol/L浓度的化合物N-414处理SGC7901和BGC823两种胃癌细胞,应用Transwell小室检测其对上述两种细胞侵袭能力的抑制作用,Western Blot检测化合物对金属基质蛋白酶2(MMP2)表达及丝切蛋白(Cofilin)磷酸化水平的影响。结果 化合物N-414对于SGC7901和BGC823两种胃癌细胞的增殖能力无明显影响,却能够以剂量依赖的方式抑制上述细胞的侵袭。Western Blot结果显示N-414能够下调胃癌细胞BGC823中MMP2的表达及Cofilin的磷酸化水平。结论 化合物N-414可能通过下调细胞中MMP2的表达及Cofilin的磷酸化水平,抑制SGC7901和 BGC823胃癌细胞的侵袭。
Objective To study the effect and mechanism of N-414 on the proliferation and invasion in SGC7901 and BGC823 cells. Methods MTT was used to detect the effect of N-414 on the proliferation of SGC7901 and BGC823 cells. The test of Transwell was used to check the invasion of SGC7901 and BGC823 cells with 0, 20, 40, 60μmol/L N-414. Western Blotting assay was adopted to identify the effect of N-414 on the expression of Matrix Metalloproteinase 2 (MMP2) and phosphorylated Cofilin. Results N-414 did not affect the proliferation of SGC7901 and BGC823 cells. But it could significantly inhibit the invasion ability of SGC7901 and BGC823 cells in a dose-dependent manner. Furthermore, N-414 could obviously down-regulate the expression levels of MMP2 and phosphorylated Cofilin. Conclusion N-414 could inhibit the invasion capacity of SGC7901 and BGC823 cells by down-regulating the expression levels of MMP2 and phosphorylated Cofilin.
