o observe the effect of Mer receptor tyrosine kinase (MerTK) on the phagocytosis of Staphylococcus aureus(S. aureus)in macrophages. Stimulating RAW264.7 macrophages with S. aureus triggered the activation of a variety of receptor molecules on the surface of macrophages,including TLR2 signaling pathway (MyD88, JNK, ERK and p38, p65),SR-A and MerTK. At the same time, Stimulating RAW264.7 cells with S. aureus also induced the activation of proteins FAK and Rac1 related to phagocytosis. Moreover, RAW264.7 cells with S. aureus led to the activation of these proteins in a time-dependent manner. Additionally, pretreatment with the specific Mer-blocking antibody significantly inhibited the activation of of MerTK in S. aureus-stimulated macrophages. However, this antibody had little effect on the activation of FAK and Rac1 in S. aureus-stimulated macrophages. Meanwhile, the effect of targeting MerTK on the phagocytosis of S. aureus in RAW264.7 cells was not significant.Thus, while MerTK negatively regulates lipoteichoic acid(LTA)-induced inflammatory response,it is not necessary for macrophage phagocytosis of S. aureus.
