雷公藤细胞固定化细胞的生长特征及甲素合成

采用海藻酸钠凝胶包埋法首次构建了雷公藤细胞的固定化培养体系,研究了固定化细胞的生长及雷公藤甲素合成的变化情况。结果表明,雷公藤固定化细胞经历了缓慢增长、指数增长及稳定增长三个生长阶段;固定化细胞培养液总糖、可溶性蛋白质及丙二醛的含量整体上随着培养时间而逐渐减少;pH值则呈现先上升后下降的变化趋势;而细胞活力却呈现先下降后上升的趋势;培养过程中POD的变化呈现"V"字型,并在培养第6天时酶活性达到最低值;SOD酶活性变化基本不大,呈现出一条比较平滑的曲线;PAL酶活性则具先急剧上升,而又缓慢下降的特征;雷公藤甲素则呈较好的同增效应,细胞内外的甲素含量都是在培养的第15天达到了分别为64.04μg/g和4.91mg/l的最大生成数量。

In this study, Tripterygium wilfordii cells embedded in Sodium alginate was used to construct the system of immobilized cell culture for study the relationship between cell culture and tirpolide compound. The results showed that there existed 3 growth stages in cell culture, namely slow growth stage, exponential growth stage, and steady growth stage. Meanwhile, the contents of total sugar, rapeseed meal protein, MDA decreased as culture time increased. However, PH value first increased and then decreased, and cell viability first decrease and then increased. In addition, in the process of culture, the change of POD showed a curve of "V" with the lowest value in sixth day, the change of SOD showed a smooth curve, and the change of PAL enzyme showed a trend that it first increased sharply and then decreased slowly. While the content of triptolide increased with the culture time and reach a maximum yield of 66.19 μ g/g and 4.91 mg/L inside and outside the cells.
