中枢神经系统固有细胞对淋巴细胞的抗原提呈作用的研究

多发性硬化(MS)的发病机制至今仍然不是很清楚。目前认为MS是中枢神经系统感染引起的自身免疫病,而在中枢神经系统中,它的固有免疫细胞与浸润的淋巴细胞之间存在一定相互作用。在EAE发病过程中,中枢神经系统固有细胞--星形胶质细胞对MOG35-55特异性淋巴细胞有抑制作用。在本次研究中我们发现,用IFN-γ预处理后的星形胶质细胞能够促进MOG35-55特异性淋巴细胞的增殖和分泌水平,这与大量MHC-II在星形胶质细胞上表达相一致。定量聚合酶联反应(qPCR)结果表明脊髓中产生IFN-γ在高峰期达到最高值。定量分析MHC-II在脊髓中的表达表明,MHC-II的表达与IFN-γ的产生是正相关。此外,在脊髓中星形胶质细胞上MHC-II的表达水平与IFN-γ的产生是正相关的。这些发现表明星形胶质细胞对EAE有促进作用。并且还发现在脊髓中IFN-γ水平增高则会使星形胶质细胞转换为抗原提呈细胞。这种转换可能会促进病理损伤进展并导致EAE的严重程度达到高峰。
he pathogenic mechanisms of multiple sclerosis (MS) is still unclear. Presently, MS is considered as autoimmune disease caused by central nervous system(CNS) infections. There is interaction between resident cells of the CNS and infiltrating lymphocytes. During the development of experimental autoimmune encephalitomyelitis (EAE), the astrocytes,which is considered as the resident cells of the central nervous system, could inhibit MOG35-55-specific lymphocytes. However, when astrocytes were pretreated with IFN-γ, they could promote the proliferation and secretion levels of MOG35-55-specific lymphocytes, coinciding with apparent expression of major histocompatibility complex (MHC)-II on astrocytes themselves. Quantitative polymerase chain reaction (qPCR) demonstrated that production of IFN-γ in the spinal cord was highest during the peak phase. Quantitative analysis of MHC-II expression in the spinal cord showed that there was a positive correlation between MHC-II expression and IFN-γ production. In addition, astrocyte MHC-II expression levels correlated positively with IFN-γ production in the spinal cord. These findings suggested that astrocytes might function as promoters of EAE. Then, in the presence of higher IFN-γ levels in the spinal cord, astrocytes were converted into antigen-presenting cells. This conversion might promote the progression of pathological damage and result in a peak of EAE severity.
