体外暴露全氟辛烷磺酸(PFOS)对淋巴细胞增值功能的影响
Effect of Perfluorooctane Sulfonate (PFOS) Exposure in Vitro on lymphocyte Proliferation Function
目的 探讨全氟辛烷磺酸(Perfluorooctane sulfonate, PFOS)体外暴露对T淋巴细胞和B淋巴细胞的增值功能影响,为进一步揭示PFOS的免疫毒作用机制提供科学依据。方法 分别以不同浓度的PFOS(0、1、5、10、50、200、300μM)处理C57BL/6小鼠脾脏淋巴细胞,培养48h后,以刀豆蛋白A(ConA) 和大肠杆菌脂多糖(LPS)作为刺激源,采用MTT法检测T、B淋巴细胞增殖功能。结果 淋巴细胞的增值功能与PFOS暴露剂量并不存在剂量-反应关系。300μM PFOS暴露组T淋巴细胞的增值功能显著的低于对照组(p<0.05);而B淋巴细胞增值功能暴露于200μM PFOS时显著的受到抑制(p<0.05)。结论 PFOS体外暴露可降低淋巴细胞的增值功能,且B淋巴细胞对PFOS的敏感性要高于T淋巴细胞。
Objective To assess the effects of lymphocyte proliferation function by perfluorooctane sulfonate (PFOS) exposure in vitro which provide scientific evidence for further revealing immunotoxicity mechanism of PFOS. Methods Splenocytes from C57BL/6 mice were treated with PFOS at different concentration in vitro (0, 1, 5, 10, 50,200, 300μM), and then after 48 hours, the proliferation ability of the T and B lymphocytes was determined by MTT stain assay with mitogen concanavalin (Con) A or lipopolysaccharide (LPS) stimulating, respectively. Results The results showed that the T lymphocytes proliferation function was decreased significantly in 300μM PFOS group comparison with to control group, and the B lymphocytes proliferation function was decreased significantly began in 200μM PFOS group comparison with to control group. Conclusion This study indicates that PFOS exposure in vitro may decreases the lymphocyte proliferation function, and B lymphocytes may be more sensitive to PFOS than T lymphocytes.
