奥奈达希瓦氏菌MR-1葡萄糖利用突变株驯化、筛选及其产电性能研究
Screening of Glucose Utilization Mutants of Shewanella oneidensis MR-1 and the Electrochemical Performance
奥奈达希瓦氏菌MR-1是产电模式菌;葡萄糖是自然界常见、含量丰富的电子供体。据报道,奥奈达希瓦氏菌 MR-1不能利用葡萄糖生长,限制了其在以葡萄糖为底物的MFC中的应用。本文以葡萄糖为生长底物来驯化奥奈达希瓦氏菌MR-1,并将驯化菌应用到以葡萄糖为唯一电子供体的MFC中,并进一步探讨其对MFC产电性能的影响。结果表明:驯化菌能够利用葡萄糖生长并产电,驯化菌在葡萄糖浓度为5g/L的MFC中最优输出电压可达424.7 mV,其功率密度达到137.3 mW/m2 ,两者分别是原始菌的4.2倍和19.1倍。表明驯化菌在能够利用葡萄糖生长之后,使其以葡萄糖为电子供体的产电水平也大幅度提升。
Shewanella oneidensis MR-1 is a model exoelectrogenic microorganism. Glucose is generally considered as a common and abundant electron donor in nature. S. Oneidensis MR-1 was reported to be unable to use glucose to grow, which limiting its use in the glucose feeding MFC. In this study, glucose was used as a substrate to domesticate S. oneidensis MR-1. And mutants capable of glucose utilization were sceened out. Then, the electrochemical performance of these mutants were investigated in glucose feeding MFCs. The results showed that domesticated mutants can use glucose to grow and generate electricity. When the glucose concentration reached 5g/L in MFC, the optimal output voltage of the mutant-inoculated MFC is up to 424.7 mV and its power density reached 137.3 mW/m2, which is 4.2 folds and 19.1 folds of that obtained from MFC inoculated with the wild-type strain, respectively.
