兔膝关节骨组织石蜡切片的制备
Preparation of articular joint tissue section of rabbit knee
目的:探索一种简单、快速的制作兔膝关节骨组织石蜡切片的方法,观察关节软骨和骨组织微观结构,为制备高质量骨组织石蜡切片和组织病理学研究提供实验基础。方法:在兔一侧膝关节的股骨内侧髁关节面制造直径3 mm深度4 mm的全层关节软骨缺损。术后4周,分离股骨髁,经过10%中性甲醛固定和混合酸脱钙后制备骨组织石蜡切片,并行苏木精-伊红(HE)和番红-固绿染色。结果:正常侧,透明软骨细胞规则排列、层次清晰,潮线和软骨下骨清晰可辨;造模侧,损伤填充区细胞表现为软骨样细胞特征。结论:该法制备的骨组织切片,周期短、标本平坦完整,软骨和骨组织结构分明、细胞和基质染色清晰,是对传统方法的有效改良。
Objective To aid at the histological evaluation of hyaline cartilage and bone, we provide a simple and rapid method of tissue preparation for articular joint histology. Methods A defect 3 mm in diameter and 4 mm in depth, penetrating the subchondral bone plate, was created on the medial femoral condyle of rabbit knee using a stainless-steel electric drill. The entire femoral condyles of the knees at 4 weeks after surgery were used to develop the tissue section technique. The tissues were fixed in 10% neutral-buffered formalin for 2 days, and decalcified in the solution containing 7% w/v AlCl3.6H2O, 5% Formic acid and 8.5% HCl for 7 days, and embedded in paraffin. Serial 5 μm sections were stained by hematoxylin-eosin (HE) and safranin-O and examined microscopically. Results Histological evaluation of the tissue sections revealed the morphological preservation of bone and cartilage cellular structures with proper HE staining characters in both repair and adjacent areas of femoral condyle. Conclusion The technique provided a reliable method for preparation of paraffin section of articular joint, which should facilitate the histological evaluation of hyaline cartilage of articular joint and bone.
