目的:探讨针刺对2型糖尿病大鼠骨骼肌细胞GLUT4基因表达的影响及针刺与优降糖对2型糖尿病大鼠骨骼肌细胞GLUT4基因表达的比较。方法:SD雄性大鼠100只,经高脂饲料喂养成肥胖大鼠(50只),腹腔小剂量注射链脲佐菌素(25mg/kg)造模成糖尿病大鼠(18只),随机分为针刺组、药物组(优降糖)和模型组,处理4周后,用快速血糖仪检测空腹血糖(FBS)、用放免法检测空腹胰岛素(FINS)、用原位杂交检测股四头肌细胞GLUT4基因表达,并与正常组大鼠进行对照。结果:针刺组和药物组FBS水平比模型组显著降低(P<0.05),接近正常组水平(P>0.05);针刺组FINS明显低于模型组(P<0.05),接近正常组水平(P>0.05),药物组FINS水平亦低于模型组,但无显著差异(P>0.05);针刺组和药物组的GLUT4mRNA表达明显高于模型组(P<0.01),但均未达正常组水平(P<0.01);针刺组GLUT4mRNA表达高于药物组(P<0.01)。结论:针刺和优降糖均可改善2型糖尿病大鼠骨骼肌细胞GLUT4基因的表达,针刺对2型糖尿病大鼠骨骼肌细胞GLUT4基因表达的上调作用优于优降糖。

Objective: To research the effects of acupuncture on the expression of skeleton muscle GLUT4mRNA in type 2 diabetes mellitus(T2DM) model rats and the comparison of regulating action on expression of skeleton muscle GLUT4mRNA in T2DM rats between acupuncture and Glyburide. Method: 50 of 100 SD male rats fed with high fat feedstuff became fat rats, which were injected with Streptozotocin(25mg/kg) to make T2DM model. Then T2DM rats(18) were Stochastically devided into Glyburide group(6), acupuncture group(6) and model group(6). After treatment for 4 weeks, fast blood sugar(FBS) was examined by active blood sugar meter and fast insulin(FINS) was examined by radioimmunoassav(RIA) and expression of skeleton muscle GLUT4mRNA was examined by In Situ Hybridization(ISH), the results were compared with normal rats(6). Result: After treatment, the level of FBS in acupuncture and Glyburide group are obviously lower than model group(P<0.05); the level of FINS in acupuncture is markedly lower than model group(P<0.05), the level of FINS in Glyburide group is also lower than model group but there has no statistical meaning between them(P>0.05); expression of GLUT4mRNA in acupuncture and Glyburide group are obviously higher than model group(P<0.01); expression of GLUT4mRNA in Glyburide group is lower than acupuncture group(P<0.01). Conclusion: Acupuncture and Glyburide also can improve the expression of skeleton muscle GLUT4mRNA in T2DM rats, but acupuncture’s effect is better than Glyburide.
