多浪羊MHC-DRB1基因多态性与包虫病抗性分析
The Study of The Polymorphism of MHC-DRB1 Gene in Dulang Sheep and Hydatidosis Associations
通过PCR扩增122只包虫病(细粒棘球蚴病)阴性和70只包虫病阳性多浪羊的MHC -DRB1第二外显子,产物经SacI、Hin1I和HaeⅢ三种限制性内切酶进行RFLP多态性分析。结果表明,多浪羊的MHC-DRB1基因第二外显子在Sac I、Hin1I和HaeIII酶切位点存在丰富的多态性,检测出了2、2和6种等位基因,出现了3、3和18种基因型。将包虫病阴性和阳性多浪羊的等位基因频率和基因型频率分别进行比较分析,发现等位基因HaeIII a对包虫病感染具有一定的易感性(P﹤0.05),SacI ab和HaeIII cf两个基因型对包虫病具有一定的抗性(P﹤0.05),SacI bb, Hin1I ab和HaeIII aa三个基因型对包虫病具有一定的易感性(P﹤0.05),而HaeIII be和HaeIII ef这两个基因型对包虫病具有较强的易感性(P<0.01)。

The ovine major histocompatibility complex classⅡ DRB1 second exon was amplified by polymerase chain reaction(PCR)from 122 healthy and 70 infection with Hydatidosis Duolang sheep. PCR products were chara- cterized by the RFLP technique using three restriction enzymes, SacI, Hin1I and HaeIII. The results revealed extensive polymorphisms 2 ,2 and 6 RFLP patterns were found with enzymes SacI,Hin1I and HaeIII respectively , so 3,3 and 18 kinds of genotypes were identified in Duolang sheep. By analyzing alleles and genotypic frequencies between healthy and infection with hydatidosis Duolang sheep, We found that allele HaeIII a was associated with susceptibility(P﹤0.05), and genotype SacI ab and HaeIII cf were associated with resistance(P﹤0.05), but the genotypes of SacI bb, Hin1I ab and HaeIII aa were associated with susceptibility(P﹤0.05), especially the genotypes of HaeIII be and HaeIII ef (P﹤0.01).
