目的:研究大黄素(1,3,8-trihydroxy-6-methyl-anthraquinone,emodin, Em)对3T3-L1前脂肪细胞增殖分化的影响,并探讨其作用机制。方法:体外培养3T3-L1前脂肪细胞,采用噻唑蓝法(MTT)研究大黄素(5、10、20、40、80、160μM)对3T3-L1前脂肪细胞增殖活性的影响;大黄素(20、40、80μM)干预3T3-L1脂肪细胞诱导分化,油红O染色法分析大黄素对3T3-L1脂肪细胞分化过程中胞浆脂滴积累的影响;生化法检测大黄素对3T3-L1脂肪细胞中甘油三酯(Triglyceride,TG)的影响;实时荧光PCR检测过氧化物酶体增殖物激活受体γ(Peroxisome Proliferator Activated Receptor Gamma, PPARγ)、CCAAT增强子结合蛋白(CCAAT/enhancer binding protein, C/EBPα)、脂肪酸合成酶(Fatty acid synthetase, FAS)、脂肪酸结合蛋白(adipocyte fatty acid binding protein, aP2)mRNA的表达水平。结果:大黄素能够不同程度抑制3T3-L1前脂肪的增殖;大黄素20、40、80μM均显著抑制3T3-L1前脂肪细胞的诱导分化,降低细胞中脂滴的积累;同时显著降低3T3-L1脂肪细胞中TG含量;与脂肪细胞比,大黄素40、80μM能够降低3T3-L1细胞中脂肪分化相关基因PPARγ、C/EBPα、FAS和aP2 mRNA的表达。结论:大黄素能够抑制3T3-L1脂肪细胞的诱导分化,降低细胞内TG含量,其作用机制可能与下调脂肪分化相关基因PPARγ、C/EBPα、FAS和aP2 mRNA的表达有关。

im To explore the effect and mechanism of emodin (Em) on proliferation and differentiation of murine 3T3-L1 preadipocytes, and investigate its possible mechanism. Methods 3T3-L1 preadipocytes were cultured and treated with Em different dosages. Cell proliferation was analyzed by MTT method. Oil red O staining method and spectrophotography were applied to analyze the degree of differentiation. Real-time PCR was applied to detect the mRNA expression of peroxisome proliferator activated receptor γ(PPARγ) and CCTTA enhancer binding protein α(C/EBPα). Result Em could inhibit the proliferation of 3T3-L1 preadipocytes in different degrees. Em significantly inhibited the differentiation of 3T3-L1 preadipocytes and decreased the accumulation of lipid droplets in 20, 40 and 80 μM groups. The expression of PPARγ, C/EBPα, FAS and aP2 mRNA in adipocytes of 3T3-L1 cells were significantly decreased by Em 40 and 80μM compared with adipocytes. Conclusion Em could inhibit the differentiation of 3T3-L1 adipocytes and decrease the intracellular TG content. The mechanism might be related to down-regulating the expression of adipose differentiation-related genes PPARγ, C/EBPα, FAS and aP2 mRNA expression.
