当归有抗癌、抗氧化、保护心血管等药理作用,阿魏酸松柏酯(CF)和藁本内酯(Lig)是当归的主要生物活性成分,所以高效同时提取CF和Lig具有重要意义。双水相体系(ATPS)和三液相体系(TLPS)提取的提取率高、选择性好。ATPS和TLPS均可用于多组分提取,但ATPS和TLPS的差异及如何选择成为困扰人们的问题。本文采用乙醇-NaH2PO4-H2O ATPS和环己烷-乙醇-NaH2PO4-H2O TLPS同时提取CF和Lig,比较两种提取体系的差异。优化的TLPS条件为:NaH2PO4溶液(3.75mol/L,pH=5.00)、乙醇和环己烷体积分数分别为34.0%、38.0%和28.0%,提取温度为45.0℃,提取时间为2.00h。ATPS的最佳提取条件:NaH2PO4溶液(3.33mol/L,pH=5.00)和乙醇体积分数分别为47.8%和52.2%,提取温度为40.0℃,提取时间为1.00h。ATPS与TLPS相比,CF产率提高了28.9%,Lig产率提高了11.4%。ATPS产率比TLPS产率高的原因有待进一步研究。

ngelica sinensis (Oliv.) Diels exhibits a variety of pharmacological activities, including anti-cancer, anti-oxidation, cardiovascular protection and others. Coniferyl ferulate (CF) and ligustilide (Lig) are the main bioactive components of Angelica sinensis. Therefore, efficient extraction and separation of CF and Lig is of great significance. Newly rising aqueous two-phase system (ATPS) and three-liquid-phase system (TLPS) have the advantages of high extraction yield and good selectivity. Both ATPS and TLPS could be used in extraction of multi-components, but choice of ATPS or TLPS is a question for researchers. In this study, CF and Lig were simultaneously extracted by ethanol-NaH2PO4-H2O ATPS and cyclohexane-ethanol-NaH2PO4-H2O TLPS, and the extraction effect were compared between ATPS and TLPS. The optimal extraction conditions of TLPS were as follows: NaH2PO4 solution (3.75mol/L, pH=5.00), ethanol and cyclohexane volume fractions were 34.0%, 38.0% and 28.0%, respectively, extraction temperature was 45.0℃, extraction time was 2.00 h. For the extraction at 40.0℃ for 1.00 h with optimal NaH2PO4 solution (3.33mol/L, pH=5.00) volume fraction 47.8%. Compared to those with TLPS, the CF yield with ATPS and Lig yield with ATPS increased by 28.9% and 11.4% respectively. The reasons for good yields of ATPS will be further investigated.
