由补阳还五汤提出的两类有效组分对凝血酶诱导血管内皮细 胞tPA和PAI-1的释放和TF、TFPI、TM基因表达的影响
Effects of two active fractions extracted from Buyang Huanwu Decoction on the release of tPA and PAI-1 and the expressions of TF, TFPI and TM mRNA in cultured vascular endothelial cells induced by thrombin
Objective To observe the effects of two active fractions extract from Buyang Huanwu Decoction(BHD) on the release of tPA and PAI-1, the expressions of TF, TFPI and TM. in cultured vascular endothelial cell (VEC) induced by thrombin. Methods The endothelial cells were cultured in the medium including thrombin and drugs, the LDH activity, the level of tPA and PAI-1 and the expressions of TF, TFPI and TM mRNA were measured afer 24 hours. Results ①After ECV-304 were stimulated by thrombin, LDH activity was increased obviously(P<0.05), the release of tPA was increased evidently(P<0.01), the expressions of TF and TFPI mRNA were inhanced(P<0.05), but the release of PAI-1 and the expressions of TM mRNA had no significant change. ②BHD, Alkaloid and Glycosid have no significant effect on the increasement of LDH activity induced by thrombin.③BHD could decrease the release of tPA(P<0.01);Alkaloid has no significant effect on the release of tPA. Glycoside(1.25mg/ml) could increase the release of tPA(P<0.05), but Glycoside(2. 5mg/ml) has no significant effect. ④Alkaloid and BHD could reduce the release of PAI-1 of ECV-304, Glycoside(5mg/ml) has the same effect. ⑤Alkaloid, Glycoside and BHD could reduce the expression of TF mRNA induced bu thrombin. Alkaloid(0.5,1mg/ml) could reduce the expression of TFPI mRNA. ⑥All drugs have no sifnificant effect on the expression of TM mRNA. Conclusion Thrombin has hameful effect on the ECV-304, it could induce antithrombotic change of VEC. BHD, Alkaloid and Glycoside could adjust the change of anticoagulant and fibrinolytic function of VEC to normal. It suggest that Alkaloid and Glycoside may be a pharmic base of antithrombosis of BHD.
