Research progress of the differentiation of human pluripotent stem cells towards human pluripotent stem cells in vitro

间充质干细胞是组织工程种子细胞之一,已广泛应用于基础研究和临床研究,但存在扩增能力有限和容易老化等问题。人多能干细胞(human pluripotent stem cells, hPSCs)具有独特体外无限自我更新和分化潜能,是目前组织工程和细胞治疗最有前景的种子细胞来源。近年来研究发现人多能干细胞来源间充质干细胞(human pluripotent stem cells derived mesenchymal stem cells, hPSCs-MSCs)具有与成体间充质干细胞类似的分化功能,能应用在各个领域,相较成体间充质干细胞还具有体外增殖能力强、来源丰富、不存在免疫原性风险等优点。但同时,它也存在成分不明确、诱导效率低、鉴定标准不明确等问题需要研究人员去探索解决。文章将对hPSCs-MSCs的分化、鉴定和纯化方法进行综述,指出其目前应用存在的问题和不足,并展望其应用前景,以期促进hPSCs-MSCs的临床应用。

Mesenchymal stem cells, as one of the seed cells for tissue engineering, have been widely used in various aspects like autoimmune, tumor treatment, injuries and so on. However, a number of factors such as inadequate numbers of cells obtained from tissue biopsies, limited extension times and impaired growth and function. Additionally, the differentiation ability of MSCs are impaired by aging and aging-related diseases. Because of the unlimited self-renewal and multi-differentiation potential in vitro, human pluripotent stem cells (hPSCs) are becoming the most promising source of seed cells for tissue engineering and cell therapy. Recently, many researchers have found that human pluripotent stem cells derived mesenchymal stem cells (hPSCs-MSCs) have similar properties to adult mesenchymal stem cells. Compared with adult mesenchymal stem cells, hPSCs-MSCs also have the advantages of stronger proliferation ability, unlimited cell number, and lower immunogenic risk. But at the same time, it also has problems such as unclear components, low induction efficiency, and unclear identification standards, which need to be explored and solved by researchers. The differentiation, identification and purification methods of hPSCs-MSCs was reviewed, and we also point out the problems and deficiencies in current applications, and prospect their application, in order to promote the clinical application of hPSCs-MSCs.
