水稻OsTPR1蛋白与乙烯受体蛋白OsERS1相互作用的酵母双杂交技术检测

OsTPR1是水稻TPR蛋白家族的重要成员之一,可能参与了乙烯信号转导。本文构建了酵母共表达载体pGBKT7-OsTPR1和pGADT7-OsERS1、pGADT7-OsERS1(952-1359)、pGADT7-OsERS1(1360-1908)载体,共转酵母AH109。检测结果表明,OsTPR1与OsERS1具有相互作用,且关键域为OsERS1(952-1359)和OsERS1(1360-1908),这为深入探讨OsTPR1蛋白在水稻乙烯应答反应中的作用及分子基础提供了依据。
OsTPR1 was a important member of TPR proteins in rice, may participate in the ethylene signal transduction. Theyeast expression vector pGBKT7-OsTPR1 and pGADT7-OsERS1, pGADT7-OsERS1(952-1359), pGADT7-OsERS1(1360-1908) were constructed successfully, and then co-transformed into AH109 yeast cells. The yeast two-hybird system detected the interaction between OsTPR1 and OsERS1. The OsERS1 domain OsERS1(952-1359) and OsERS1(1360-1908) were identified to play key role in the intereaction between OsTPR1 and OsERS1. OsTPR1 participated in ethylene signaling transduction pathway associated with OsERS1.
