印记基因对哺乳动物胚胎发育和出生后的生长具有重要的调节作用,包括肿瘤在内的许多疾病与印记基因的异常表达有关。因此,对基因组印记展开研究,无论是对于发育和表观遗传学领域基础理论的发展,还是对于有关疾病的诊断与治疗都具有重要意义。基因组印记研究的首要任务是鉴定新的印记基因并解析其表达模式。因此,本研究根据印记基因成簇分布的特点,将小鼠1号染色体Zdbf2-Gpr1印记区内的EST CJ051953作为侯选印记基因,利用基于SNP标记的RT-PCR产物直接测序法对该EST进行印记分析,以期鉴定新的印记基因,为小鼠印记基因家族增加新成员。同时,利用原位杂交技术,对该EST进行表达分析,为其后续表达调控及功能研究奠定基础。印记分析结果表明,EST CJ051953优先表达父本等位基因,表现为不完全印记,具有印记区边界基因的印记特征。表达分析结果表明,该EST在小鼠神经系统中表达,预示它们可能与小鼠神经系统的分化和发育相关。

In mammals in particular, imprinted genes play important roles in fetal growth, development and maternal behavior. Abnormal expression of imprinted genes is implicated in many diseases including cancer. Therefore, a study of genomic imprinting is interesting for not only development of the theory of developmental biology and epigenetics, also treatment and diagnosis of related diseases. The primary task of the study of genomic imprinting is the identification of new imprinted genes and analysis of their expression pattern. Therefore, the study analysed imprinting status of an EST CJ051953 in Zdbf2-Gpr1 imprinting domain on chromosome 1 as candidate imprinted genes based on clusters features of imprinted genes by direct sequencing of SNPs approach. Fothermore, the study analysed expression patterns of the candidate imprinted genes by in situ hybridization (ISH). Imprinting analysis showed preferential paternally expressed of CJ051953 in the mouse. Expression analysis showed that CJ051953 was highly and continuously expressed in nervous system, which suggested that the EST may be involved in the mouse embryonic nervous system development.
