目的 研究乙醇代谢酶基因多态性与酒精性肝硬化(alcoholic liver cirrhosis, ALC)的关系,探讨遗传因素对其易感性的影响。 方法 应用PCR-RFLP法检测汉族男性健康者、酗酒者及ALC患者中ADH3、ALDH2和CYP2E1基因型并比较其基因频率。 结果 ALDH2两种等位基因在对照组、酗酒组及ALC组中的频率具有显著差异,ADH3及CYP2E1的两种等位基因在三组间的频率差异则均无显著性。综合ADH3、ALDH2和CYP2E1基因多态性的影响,发现ALDH2*1等位基因在酗酒组及ALC组中的频率显著高于该等位基因在对照组中的频率,而ALDH2*2等位基因在酗酒组中的频率则显著低于它在ALC组中的频率。 结论 ADH3及CYP2E1基因多态性与ALC的发生无关,等位基因ALDH2*1是汉族男性酗酒的诱因,等位基因ALDH2*2则提高其携带者ALC的患病风险。

Objectives To study the relationship between polymorphisms of alcohol-metabolizing enzymes and alcoholic cirrhosis (ALC), and investigate the influence of genetic factors on the susceptibilitys to it. Methodes PCR-RFLP was used to determine the genotypes and alleles frequences of ADH3, ALDH2 and CYP2E1 of healthy controls , alcoholics without evidence of liver disease and patients with ALC in male Han Chinese. Results The frequencies of the two alleles of ALDH2 in the controls, alcoholics and ALC were significantly different, while the frequencies of the two alleles of ADH3 and CYP2E1 in the three groups were similar. Combined analysis of ADH3, ALDH2 and CYP2E1 revealed that the frequencies of the allele ALDH2*1 in alcoholics and ALC patients are much higher than that one in the controls, and the frequencies of the allele ALDH2*2 are remarkably lower in alcoholics than in ALC group. Conclusions There is no association between polymorphisms of ADH3 and CYP2E1 and ALC, the allele ALDH2*1 is an inducement for the developing of alcoholics while the allele ALDH2*2 is related with the increase risk of ALC .
