从本实验室构建的cDNA文库中查找到一条阅读框为273bp的,编码90个氨基酸残基的基因序列,经氨基酸同源性比对发现其为蚕素Bbx-b4基因序列。与胰岛素在结构和功能上都具有很大的同源性,由信号肽、B链、C肽和A链组成。天然蚕素切除信号肽和C肽,由A链和B链组成,A链和B链之间以二硫键相连,分子量约为4.0kDa。将Bbx-b4基因与家蚕杆状病毒多角体蛋白Polh基因融合,克隆到表达载体pET-28a(+)上,在大肠杆菌BL21(DE3)中融合表达,利用多角体特点,通过调节缓冲液PH溶解包涵体和镍柱亲和层析的方法进行蛋白初步纯化,将纯化的重组蛋白免疫新西兰雄兔制备多克隆抗体;同时将Bbx-b4基因克隆到pFastBac1中,构建重组质粒,在家蚕细胞BmN中表达。Tricine-SDS-PAGE和Western blotting检测Bbx-b4在家蚕细胞BmN和蚕蛹中的表达情况。流式细胞分析细胞周期实验表明Bbx-b4具有促进细胞生长的功能;四氧嘧啶高血糖小鼠降血糖实验表明家蚕中表达的Bbx-b4具有一定的降血糖活性,但是较正常蚕蛹活性不显著。

We found the Bbx-b4 gene from the cDNA library constructed by our laboratory. After Blasten based on NCBI database and alignment of deduced amino acid sequence homology,it was found to be the Bbx-b4 gene.Its open reading frame(ORF)is 273 bp and encod 90 amino acids . Bombyxin and human insulin are so similar that they have the same tertiary structure. Its molecular weight is about 4.0kDa. We cloned the fusion gene of Bbx-b4 and gene Polh(polyhedrin,Polh) into the vector pET-28a and pFastBac1 to construct recombinant plasmid, and the fusion protein was expressed by E.coil system and Bombyx mori NPV expressing system, respectively. The fusion protein expressed in the E.coil syste remained as inclusion body. According to the principle of the alkali fusion of Polh protein,we purified the fusion protein by dissolving inclusion protein in different PH buffer solution and metal-chelating affinity chromatography.The purified fusion protein was used to immunize male New Zealand rabbit. Western blot analysis revealed that the protein was expressed in siliworm cell and fresh silkworm pupa successfuliy.Flow cytometry cell cycle experiment proved the Bbx-b4 has the function of promoting the growth of cells. The expressed protein in fresh silkworm pupa could obviously reduce the tall fasting glucose in the mice whice were injected by ALX(Alloxan tetrahydrat),but its effect is not obviously comared by the natural silkworm pupa.
