重金属胁迫对水稻粳籼亚种特定序列DNA甲基化的影响
he Influence of DNA Methylation of Specific Sequences in Rice Two Subspecies Induced by Heavy Metal Stress
植物在重金属胁迫下会对体内关键的生理过程进行相应的调控,如光合作用、呼吸作用、抗氧化系统和激素代谢等。但近些年越来越多的研究表明表观遗传参与到植物的各种胁迫应答中,然而对于其具体的抗逆机制研究的还不清晰,深入研究表观遗传参与到植物抗逆的机制对于进一步将其应用到植物育种和生物保护都是必需的。本实验选择了水稻两个亚种粳稻品种"日本晴"和籼稻品种"9311"为实验材料,对其进行重金属处理,用Southern杂交的方法检测在重金属胁迫后水稻中特定的序列包括转座子和低拷贝基因的甲基化变化,结果表明水稻粳稻品种"日本晴"发生了特异的去甲基化变异,并且变异主要是发生在CHG位点,而籼稻品种"9311"在胁迫前后所检测的序列均保持稳定的DNA甲基化,暗示着表观遗传在水稻亚种应对重金属胁迫时所起的作用机制可能不同。
Under heavy metal stress, plants would regulate their corresponding physiological progresses such as photosynthesis, respiration, antioxidant system and hormone metabolism, etc. Recent years a growing numbers of research suggests that epigenetic regulations involved in the plant coping with different kinds of stress response, however, the detain mechanisms involved in the stress response are still not clear, some further studies show that epigenetic mechanism involves in the plants coping with stress and which is necessary for the future application in plant breeding and conservation. In this study, the japonica rice "Nipponbare" and indica rice "9311" were used as materials, then treated them with heavy metals, using Southern blot method, we detected the DNA methylation changes of special sequences including transposons and low copy cellular genes in treated rice plants compared with the Mock plants, the results showed that japonica rice "Nipponbare" occurred specific hypomethylation and predominately in CHG sites in most of the investigated sequences, while indica rice "9311" kept stable in all investigated sequences, suggesting that epigenetic mechanism involved in rice subspecies coping with heavy metal stress may be different.
