无机纳米粒子体外抑制Bel-7402肝癌细胞增殖的实验研究
Investigation on proliferation inhibited of Bel-7402 hepatoma cells by nanaparticles in vitro
目的 体外筛选具有抑制Bel-7402肝癌细胞增殖的无机纳米粒子。方法 用均相共沉淀法制备羟基磷灰石(HAP)、锶磷灰石(SrAP)和二氧化钛(TiO2)纳米粒子,应用电位粒度分析仪测定纳米粒子粒径和粒度的分布特点。采用噻唑兰(MTT)实验检测无机纳米粒子对Bel-7402肝癌细胞增殖的抑制作用。结果 HAP、SrAP、TiO2 纳米粒子对Bel-7402肝癌细胞增殖的抑制率分别是79.10±4.36%、74.81±7.13%和84.64±5.49%。同时HAP纳米粒子对Bel-7402肝癌细胞的抑制作用与其粒径的大小关系密切,平均粒径<100nm的纳米粒子对Bel-7402肝癌细胞的抑制效果较好。且不同成份的纳米粒子对Bel-7402肝癌细胞的抑制作用在一定的浓度范围内呈现时间依赖和剂量依赖效应。结论 3种不同成分的无机纳米粒子对Bel-7402肝癌细胞的增殖均有一定的抑制作用,并且粒径<100nm的HAP纳米粒子抑制作用尤为明显。
bjective To explore the inhibition of nanaparticles on Bel-7402 hepatoma cells so that could sieve the best nanaparticles to research the further mechanism. Methods Nanaparticles, such as HAP, SrAP and TiO2 were prepared by homogeneous conprecipitation and assayed by Zetasizer3000HS to analyze nanoparticles radius and characteristic of distribution. MTT is one of the experiments to test the metabolization of the cell , using this to observe the proliferation of the cancer cells after affected by nanaparticles. Result The maximum ratio of inhibition by nanaparticles, HAP, SrAP and TiO2 is 79.10±4.36%,74.81±7.13%and 84.64±5.49% respectively. At the same time there is intimate connection between inhibition effect and the radius of HAP nanoparticles. Proliferation of Bel-7402 hepatoma cells was inhibited better by HAP whose average radius in the range of 100nm .
