生物纳米膜结构exosome在疾病治疗中的研究进展

Exosome是来源于生物体的一种囊泡状结构,直径在30-100 nm,可参与细胞间信息传递,是生物体内的天然信使。Exosome最初发现于40多年前,随着对其生物形成过程和运输机理的深入了解,近年来成为研究焦点。研究发现,exosome不但与肿瘤的转移、抗原提呈等过程有密切关系,而且还可作为疾病发展的诊断指标。尤为重要的是,exosome作为一种纳米结构,可以作为药物的运输载体,为基因治疗提供了一条崭新的途径。我们课题组首先利用未成熟树突状细胞来源的exosome,通过基因修饰的手段,将靶向脑神经的短肽插入到exosome的膜上,从而赋予了exosome靶向性,使其成功地跨越血脑屏障,将药物运输到靶向组织中,介导特异性的基因下调。同样,exosome也被广泛用于肿瘤的免疫治疗及其他相关疾病的治疗研究中。本论文将侧重于讨论exosome在基因治疗和免疫治疗领域的最新进展及exosome在疾病治疗方面的应用潜力和存在的局限性
Exosomes are membrane-bound vesicles with the size of 30-100 nm, which can be shed from a variety of cells. Exosomes are important natural messengers involving in cellular communication by carrying mRNA, microRNA and protein. The discovery of exosomes can be dated back to four decades ago and they become fast-extending field recently with further understanding of their biogenesis and the mechanism of cellular transfer. It has been shown that exosomes are not only involved in the metastasis of tumors and the presentation of antigens in antigen-presenting cells but also can be used as diagnostic or prognostic markers for the progression of various diseases. Notably, exosomes show huge potential in drug delivery as biological nanoparticles and provide a novel strategy for gene therapy. Previously our group genetically modified immature dendritic cell-derived exosomes by anchoring a neuron-specific peptide on the surface of exosomes, by which we could confer the targeting property to exosomes. We demonstrated that genetically modified exosomes could successfully cross blood-brain barrier and deliver the cargo to targeted neurons and mediate the targeted gene knock-down. Furthermore, exosomes have been extensively utilized for immunotherapy in various tumors e.g. melanoma and non-small cell lung cancer and other diseases. In this review, we will focus on the recent development of exosomes in the field of gene therapy and immunotherapy. Moreover, we would like to discuss further about the potential and limitation of exosomes as a novel drug delivery system.
