目的:从JNK- caspase-3信号通路探讨苦碟子注射液治疗急性脑缺血的可能作用机制。方法:SD大鼠随机分为假手术组、模型组(R6h、R24h、R72h)和苦碟子组(K6h、K24h、K72h),取大脑缺血侧皮层,Western blot法检测JNK、p-JNK和caspase-3蛋白表达。结果:与假手术组比较,模型组p-JNK、caspase-3蛋白表达增加,在R24h达到高峰;相应时间点苦碟子注射液组p-JNK和caspase-3蛋白相对含量均减少,具有统计学意义(P<0.01)。结论:苦碟子注射液可能通过阻断JNK信号通路,抑制caspase-3蛋白表达降低细胞凋亡,从而减轻脑缺血再灌注的损伤。

im:To discuss the possible mechanism of Ku Diezi injection on JNK- caspase-3 signal pathway in the treatment of acute cerebral ischemia. Methods: Animal model of focal cerebral ischemia and reperfusion is the thread occlusion technique induced middle cerebral artery occlusion in rats. Rats were randomly divided into seven groups: sham group, 6h, 24h, 72h of reperfusion respectively after 1.5h of ischemia by transient occlusion of middle cerebral artery, and Ku Diezi injection treatment groups . Detecting the protein expression of JNK/p-JNK and caspase-3 by Western blot. Results:Compared with the sham group, p-JNK and caspase-3 protein expression are peaked in R24h of model group, corresponding time points Ku Diezi injection group reduce the p-JNK and caspase-3 protein expression, relative with statistical significance (P <0.01). Conclusion:Ku Diezi injection probably blocking the JNK signaling pathway, inhibiting caspase-3 protein expression and decreas apoptosis, thereby reducing cerebral ischemia-reperfusion injury.
