柑橘β-隐黄质的体外抗氧化活性及其对血管平滑肌细胞增殖的调控研究
Research on Antioxidant Activities in Vitro and Modulatory Effect on VSMC Proliferation of Citrus β-Cryptoxanthin
本文测定了柑橘β-隐黄质对体外自由基的清除能力,观察和分析了柑橘β-隐黄质处理对的血小板衍生生长因子(PDGF BB)诱导的血管平滑肌细胞(VSMC)增殖的影响。结果表明:β-Cry对DPPH+、ABTS+o的清除能力优于Trolox,IC50值分别为9.52μg/ml和8.26μg/ml。MTT检测结果显示:β-隐黄质处理组与正常对照组VSMC细胞增殖能力相当,相较于PDGF BB诱导组,β-隐黄质处理对PDGF BB诱导VSMC增殖具有显著抑制作用(p<0.05)。细胞周期分析表明,PDGF BB诱导组VSMC细胞G1/G0期细胞比例相比于对照组VSMC细胞下降,S期细胞比例增加(p<0.05),而β-隐黄质+PDGF BB处理组调节了因PDGF BB诱导的VSMC细胞周期比例变化(p<0.05)。Western blot分析表明,PDGF BB诱导组VSMC的Cyclin E、Cyclin D1蛋白表达水平上调,p21蛋白表达下降下调,β-隐黄质处理对此具有调节作用,与细胞周期分析相符。研究认为β-隐黄质可能通过调节细胞周期变化以及CyclinD1、CyclinE、p21蛋白的表达水平等抑制PDGF BB诱导的VSMC增殖,这一结果将为以柑橘类胡萝卜素提取物为原料的功能性活性研究提供基础。
he aims of this research were to explore the activities of β-Cryptoxanthin (β-Cry) in vitro by free radical scavenging ability assays, and anti-proliferation effect observation on vascular smooth muscle cell (VSMC). The results showed that β-Cry had highly scavenging abilities of DPPH radical and ABTS radical, and IC50 values were 9.52μg/ml and 8.26μg/ml, respectively, which were better than Trolox. MTT assay implied that β-Cry could inhibit the VSMC proliferation induced by PDGF BB in a dose effect. Cell cycle distribution of VSMC measured by flow cytomety indicated that the proportion of VSMC treated by PDGF BB of G1/G0 phase cells decreased by induction of PDGF BB, and the S phase cells increased in comparison with control group (p<0.05), which would be in consistent with the cell cycle related protein expression detected by Western blot, as Cyclin E and Cyclin D1 were up regulated, however, p21 was inhibited. In β-Cry+ PDGF BB treated group, β-Cry regulated proportion of G1/G0 phase cells and S phase cells, and the expression of Cyclin E, Cyclin D1 and p21, respectively. In conclusion, β-Cry may inhibit the PDGF BB induced VSMC proliferation via down regulating the expression of cyclin E and cyclin D1 induced by PDGF BB, and up regulating p21 expression, respectively, and this would provide fundamental results for the evaluation of citrus carotenoids bioactivities.
