目的:检测基质金属蛋白酶-2(MMP-2)、趋化因子受体CXCR4和骨形态发生蛋白-2(BMP-2)在乳腺癌患者肿瘤组织中的表达及其与临床指征的相关性,探讨它们在乳腺癌中的生物学意义。方法:选择临床及病理资料完整的乳腺癌患者103例,其中未发生淋巴结转移者55例,发生淋巴结转移者48例。应用免疫组织化学方法检测肿瘤组织中MMP-2、CXCR4和BMP-2的表达情况,并结合患者年龄、肿瘤大小、淋巴结转移、临床分期等,探讨它们之间的相互关系。结果:MMP-2、CXCR4和BMP在有淋巴结转移的乳腺癌组织中的阳性表达率分别为58.3%、91.7%和58.3%,均高于无淋巴结转移者的阳性表达率(29.1%、70.9%和38.2%),差异有统计学意义(P<0.05);MMP-2分别与CXCR4、BMP-2的阳性表达率呈正相关(r=0.162,P<0.05;r=0.316,P<0.01)。CXCR4与BMP-2的阳性表达率也呈正相关(r=0.271,P<0.05)。结论:MMP-2、CXCR4和BMP-2在有淋巴结转移的乳腺癌组织中的阳性表达率高于无淋巴结转移者,提示MMP-2、CXCR4和BMP-2共同参与乳腺癌的浸润和转移,联合检测三者表达可评估乳腺癌的转移情况。

Objective:To investigate the expression and the biological significance of matrix metalloproteinase-2(MMP-2)、chemokines receptor CXCR4 and bone morphogenetic protein-2(BMP-2)in patients with breast cancer and analyze their correlation. Methods:A total of 103 breast cancer patients including 48 cases with lymph node metastasis were analysed. The expressions of MMP-2、CXCR4 and BMP-2 were detected by inmunohistochemical staining. The correlations among the expressions of MMP-2、CXCR4、BMP-2 and clinical stage,lymph node metastasis and tumor size were analysed. Results:The positive expression rates of MMP-2、CXCR4 and BMP-2 were 58.3%、91.7% and 58.3% in the group of breast cancer with lymph node metastasis, significantly higher than those in the group of breast cancer without lymph node metastasis which were 29.1%、70.9% and 38.2% ,respectively(P<0.05);Positive correlation could be found between MMP-2 and CXCR4、BMP-2 expression(r=0.162,P<0.05;r=0.316,P<0.01)and also between CXCR4 and BMP-2 expression (r=0.271;P<0.05). Conclusion:MMP-2、CXCR4 and BMP-2 are highly expressed in the group of breast cancer with lymph node metastasis. These results suggest that they might be important factors in breast metastasis. These proteins are indicators of metastasis of breast cancer.
