基于单细胞转录组学数据的细胞通讯研究方法与应用

细胞通讯(CCC)是调节生物体生长发育、维持内环境稳态、调控免疫反应的基础生物过程,当CCC过程不能正常进行时,就会影响疾病的发生和发展。既往CCC分析方法要求很高,且只能研究特定细胞类型中少数分子的CCC。随着单细胞转录组测序(scRNA-seq)技术的发展,提出了多种基于scRNA-seq转录组数据进行CCC研究的方法,使构建更全面的细胞间通讯图谱成为可能。本文综述了近年来出现的基于scRNA-seq数据进行CCC推断的方法,并对目前评价CCC推断方法和推断结果的方法进行了总结,最后对其发展前景进行了展望。
ell-cell communication (CCC) is a basic biological process that regulates the growth and development of organisms, maintains the homeostasis of internal environment and regulates the immune response. When CCC process cannot be carried out normally, it will affect the occurrence and development of diseases. Previous CCC analysis methods were very demanding and could only study CCC of a few molecules in a specific cell type. With the development of single cell transcriptome sequencing (scRNA-seq), a variety of methods for CCC study based on scRNA-seq transcriptome data have been proposed, which makes it possible to construct a more comprehensive map of cell-cell communication. In this paper, we reviewed the methods of CCC inference based on scRNA-seq data in recent years, summarized the current methods of evaluating CCC inference methods and inference results, and finally prospected their development prospects.
