芍药苷通过p38MAPK及PKA调节氨诱导星形胶质细胞AQP4表达
Paeoniflorin Regulates AQP4 Expression of Ammonia Treated Astrocytes via p38MAPK and PKA Pathway
目的 观察芍药苷对氨处理星形胶质细胞水通道表达的影响及其相关机制。方法 原代培养星形胶质细胞用氯化铵造模后,用不同浓度芍药苷或PKA阻断剂H89处理。用western检测AQP4表达和p38MAPK磷酸化水平。结果 不同浓度的芍药苷均对模型细胞产生影响,字不同程度上降低AQP4及p38磷酸化水平。使用PKA阻断剂H89后,模型细胞的AQP4和p38MAPK磷酸化水平升高。结论 芍药苷能够调节处理星形胶质细胞水通道表达的异常,其机制与p38MAPK和PKA有关。
Objective:To observe the influence of paeoniflorin on the experssion of aquaporin 4 of ammonia treated astrocytes and the underlying mechnaism. Methods: Paeoniflorin of different concentration or H89 were administrated to primary cultured astrocytes after astrocytes were treat with ammonium chloride. Westernblot assay was used to detect the expression of AQP4 and phosphorylated p38 MAPK. Results : Paeoniflorin of different concentration can influence ammonia treated astrocytes via suppress the expression of AQP4 and phosphorylation of p38 MAPK in varying degrees. After administrated with H89, the expression of AQP4 and phosphorylation of p38 MAPK of astrocytes were elevated. Conclusion:Paeoniflorin can regulate the abnormal expression of AQP4. The underlying mechanism is related PKA and p38 MAPK.
