皮肤缺损再生过程中毛囊附属器官的缺失至今仍是重大的挑战。本文旨在研究含有0.4%胎牛血清、40ng/mL的肝细胞生长因子(HGF)和0.7μM氢化可的松的DMEM/F12培养基对诱导骨髓间充质干细胞(MSC)向类毛囊细胞分化的影响,以期为毛囊再生提供合适的种子细胞。论文分别通过细胞形态观察,实时聚合酶链式反应(RT-PCR)和免疫组织化学染色等手段,考察了诱导分化前后细胞形态和关键标志物的变化。结果显示,诱导后细胞的多个与毛囊相关的标志物的表达水平都明显高于未诱导细胞。可见在HGF与氢化可的松的协同作用下,MSC表现出向类毛囊细胞分化的倾向。

he missing of hair follicles during the skin repair and regeneration is still regarded as a major challenge. In order to induce MSC differentiate into hair follicle cells, DMEM/F12 cell culture medium with 0.4% fetal serum, 40ng/mL hepatocyte growth factor (HGF), 0.7μM hydrocortisone was investigated. After 3 weeks' induction, real-time polymerase chain reaction (RT-PCR), and immunohistochemical staining were used to detect the key markers of hair follicle cells. The results showed that the distinguished high expressions of these key markers at both gene and protein level were obtained after induction, which indicates that the synergistic function of HGF and hydrocortisone on inducing MSC into hair follicle cells.
