检测鼠粪中隐孢子虫卵囊方法的研究

目的 研究比较三种检测方法对鼠粪中微小隐孢子虫卵囊的检测效果,以便更好的为临床诊断服务。方法 应用改良抗酸染色法(MAFS)、免疫荧光单克隆技术(IFA-McAb) 检测法和PCR检测法分别对含有不同数量隐孢子虫卵囊的鼠粪样品、隐孢子虫病模型小鼠的鼠粪样品和某牛场的牛粪样品进行检测。结果 MAFS检测法、IFA-McAb检测法和PCR检测法的检测域值分别为10 g鼠粪中含有隐孢子虫卵囊2.0×104、2.0×104、2.0×102个,3种方法操作1次用的最短时间依次为35 min、60 min、3 h。三者对实验鼠粪样品的检出率分别为65.0 %、70.0 %、85.0 %,牛粪样品的检出率分别为21.21 %、18.18 %、27.27 %,3种方法检出率之间差异不显著(P>0.05)。结论 MAFS检测法简单、快速,花费较少,条件要求不高,特异性与IFA-McAb检测法相似,适合于对大量样品的检测和流行病学调查;IFA-McAb检测法、PCR检测法必须应用精密仪器,更适合隐孢子虫的鉴定;PCR检测法特异性最强。
Objective To serve the clinical diagnosis better, Comparative studying the effect of different methods on detecting C.parvum oocysts in mice and cow feces. Methods Feces with grads concentration of oocysts per 10 gram were prepared, 27 mice feces and 33 cow feces were collected, and detected by the 3 methods respectively, modified acid-fast staining(MAFS), immunofluorescence using monoclonal antibody(McAb) immunolabeling by FITC against C.parvum and polymerase chain reaction(PCR). Results The least dosages of oocysts detected using forer 3 metheds in 10 g feces and the least time spended on 3 methods at a time were 2.0×104, 2.0×104, 2.0×102 and 35 min、60min、3h respectively. Masculine ratio in mice and cow feces detected were 65.0 %、70.0 %、85.0 % and 21.21 %、18.18 %、27.27 % respectively, no difference was found among the 3 methods. Conclusion The modified acid-fast staining is operated simply and quickly, which is suited for detecting quantities of samples and spends less with lower condition。Its sensitivity is the same as the method using IFA- McAb, which is suited for identifying the Cryptosporidium oocyst type in human and animals for the use of the immunofluorescence microscope. It is the highest for PCR in the sensitivity of detecting C.parvum oocyst, but PCR apparatus must be used in identifying the oocyst type.
