本文利用Baclight染色法测定活性污泥中的活菌水平,利用荧光染色剂SYTO®9和PI (propidium iodide)对活性污泥进行染色,通过荧光分光光度计测量绿色荧光(Fg,波长500-510nm)光强来表征活性污泥中活菌水平。可以直接对稀释后的污泥悬浊液进行测量,简化了测量步骤。当污泥浓度小于100mg/L时,投加25μl染色剂/mL污泥是完全过量的;污泥浓度稀释到50mg/L,其惰性颗粒物对测量产生的影响在7%以下;超声波分散的最佳条件是:超声时间2min,功率40W。

he BacLight staining method, the reliability of which had been validated by previous experiments, was used to measure the live bacteria ratio in the activated sludge. The density of green fluorescence (Fg) was measured by fluorescence spectroscopy after the samples were stained by SYTO®9 and PI. The Fg indicated the level of live bacteria. When the sludge concentration was lower than 100mg/L, the dose of staining material was 25μl/ml sample. Diluting activated sludge until the concentration was under 50mg/L, the affection of inert floc could be ignored. The sample treated by dispersing by ultrasonic (the duration was 2 min and the power was 40W) could improve the repeatability of the method.
