MPTP诱导C57/BL6小鼠帕金森病模型的步态分析
Gait analysis of Parkinson's disease model induced by MPTP in C57/BL6 mice
目的:研究MPTP(1-Methyl-4-phenyl-1,2,3,6- tetrahydropyridine,1-甲基-4-苯基-1,2,3,6-四氢吡啶)诱导C57/BL6小鼠帕金森病模型步态,探索CatWalk评价帕金森病模型的有效指标。方法:10 - 12周,20 - 25gC57/BL6雄性小鼠随机分为正常对照组与模型组。模型组腹腔注射MPTP (30mg/kgod),同时正常对照组给予等体积生理盐水,连续7天。第7天注射完MPTP3小时后进行行为学检测,利用CatWalk步态分析系统检测小鼠动、静步态指标的变化,行为学测试完后取材,免疫组化法检测小鼠黑质致密部(Substantia nigra pars compacta, SNpc)、酪氨酸羟化酶(Tyrosine hydroxylase, TH)阳性细胞数,HPLC-ECD法检测小鼠纹状体内的多巴胺(Dopamine, DA)及其代谢产物的含量。结果:免疫组化、DA及其代谢产物的含量检测结果显示,MPTP成功的复制了帕金森病的小鼠模型,CatWalk系统分析显示MPTP导致小鼠运动速度明显降低,足部接触面积增大,悬空速度降低,步长减少,后肢左右脚间距增加,步伐的协调性发生改变。结论:MPTP致帕金森病C57/BL6小鼠模型步态较正常组有明显变化,CatWalk的参数能作为评价帕金森病小鼠模型的有效指标。
IM:In order to evaluate the probability of quantitative analysis of parkinsonian gaits in MPTP-induced mice. METHODS: Male C57/BL6 mice aged 10-12weeks, weight 20 - 25g, were randomized to receive 30 mgokg -1o d-1 MPTP or physical saline for 7days. On 7th day CatWalk analysis were applied after received MPTP3h, then killed mice for Immunohistochemical analyses and striatal levels of DA、HVA、DOPAC. RESULTS: The MPTP-induced mice were successful copy the mouse of Parkinson's disease and CatWalk-assisted gait analysis results suggest mice fromexperimental group had slower walk speed 、swing、greater print area than mice from control group. Besides that, Interlimb coordination also changed. CONCLUSION: CatWalk analysis system can commendably reflect the defect in gaits of MPTP-induced Parkinson's disease.
