南极大磷虾胰蛋白酶促进皮肤创伤愈合作用研究
Effects of trypsins from the Antarctic krill on cutaneous wound healing
目的 探讨南极大磷虾胰蛋白酶促进皮肤创伤愈合效果。方法 建立动物全层皮肤缺损创伤模型,将创面分为即粗酶组、纯化酶组、木瓜蛋白酶组和生理盐水组四组,观察创面形态学变化,评价创伤表面瘢痕、测量伤口表面积、观察组织切片、测定皮肤羟脯氨酸、表皮生长因子(EGF)和碱性成纤维细胞生长因子(bFGF)含量,比较各蛋白酶创伤愈合效果。结果 建模后第十五、二十天,粗酶组和纯化酶组瘢痕表量值、愈合率、组织切片观察结果均显著显著优于木瓜蛋白酶组和生理盐水组。建模后第十天、十五天,粗酶组、纯化酶组EGF含量和bFGF含量显著高于生理盐水组,且粗酶组显著高于纯化酶或木瓜蛋白酶组。结论 南极大磷虾粗酶和胰蛋白酶能显著促进新西兰大白兔皮肤创伤愈合,粗酶效果最优,且能缩短愈合时间。
Objective To investigate the effects of the Antarctic krill trypsins on cutaneous wound healing. Methods The animal model of full-thickness skin defect was established with mechanical trauma. Each rabbit was cut four wounds which were treated with crude tryspins, pure trypsins, papin and saline. Then pathological morphology change of the wounds were observed, the scars were rated with the Vancouver Scar Scale (VSS), the wounds area was measured, the histological change were observed with tissue sections, and the EGF and bFGF contents of wound skin tissue were essayed. Results At the fiftieth and twentieth day after modeling, the effects of the crude and pure trypsins on reducing epithelite and enhancing healing rate, were better than those of the papain and saline. At the tenth and fiftieth day after modeling, the crude and pure trypsins promoted the expression of EGF and b FGF. Conclusion The results indicated that the crude and pure trypsins from the Antarctic krill can significantly promote the cutaceous wound healing, the effects of the crude trypsins were the best, which can reduce the healing time.
