延边黄牛阉牛、公牛背最长肌差异表达基因的筛选与鉴定
Screening and identification of the differentially expressed genes in Longissimus dorsi of Yanbian steers and bulls
为筛选延边黄牛阉牛、公牛背最长肌组织差异表达基因,从基因表达水平揭示阉牛和公牛不同性状差异的遗传基础。文章应用引物退火控制技术(ACP)分别检测了3头30月龄延边黄牛阉牛、公牛背最长肌组织的mRNA表达水平变化。利用20对随机引物差异显示扩增下,共获得10条ESTs,其中8个为已知的ESTs。对NADH脱氢酶亚单位6(ND6)、β-肌球蛋白重链(MYH7)、NDUFS8(NADH dehydrogenase Fe-S protein 2)和α-肌动蛋白1(ACTA1)进行了荧光定量PCR检测,结果显示ACTA1和NDUS8 在阉牛组背最长肌中的表达量显著高于公牛组(P<0.05);ND6和MYH7 在公牛组背最长肌中的表达量显著高于阉牛组(P<0.05)。结果提示,这些基因参与延边黄牛阉割前后肉质性状差异的重要调控基因。

The expressions of ACTA1 and NDUFS8 were significantly higher in the steers (P<0.05), The expressions of the NADH dehydrogenase subunit 6 and MYH7 genes were significantly higher in the bulls (P<0.05). All the ESTs were screened by ACP system were considered as the most important genes involved in meat quality differences between steers and bulls Yanbian cattle.
