新城疫病毒感染鹅后β-防御素1.3.6基因表达量变化的研究

本研究利用RT-PCR方法,从鹅的脾脏、骨髓和法氏囊等组织器官中扩增到3个鹅AvBDs基因,分别为鹅β-防御素1(AvBD1)、鹅β-防御素3(AvBD3)和鹅β-防御素6(AvBD6)。为研究鹅机体内感染新城疫病毒后,β-防御素表达量变化情况。实验采用实时荧光定量RT-PCR法,从感染新城疫病毒后36h和72h后鹅的脑、气管、肺脏、肾脏、肝脏、腺胃、脾脏、盲肠扁桃体、哈德氏腺和法氏囊10个组织中检测目的基因的含量。结果表明,受感染后,这3个AvBD基因在不同组织中的表达量变化较大,其中AvBD1基因在骨髓中的表达量显著提高,AvBD3基因在骨髓、脾和大肠中的表达量显著提高,AvBD6基因在免疫组织中的表达量都显著提高。?????
hree novel avian β-defensin (AvBD) orthologues were isolated from spleen, bone marrow and Bursa of fabricius of geese by RT-PCR. they were goose AvBD1, goose AvBD3, and goose AvBD6 respectively.In order to investigate whether the mRNA expression of goose AvBDs, was changed in response to NDV infection.Ten tissues, namely brain, trachea, lung, kidney,liver, glandular stomach, spleen,cecal tonsil, Harderian glands, and bursa of Fabricius were collected from geese on 36 and 72 hours post infection (hpi) to measure the mRNA levels of AvBDs by the Real-time RT-PCR. The results showed that highly significant upregulation of the level of expression of AvBD3 in both bone morrow, spleen and large intestinewas observed when the geese were infected with NDV. Similarly, the level of expression of AvBD1 in bone morrow of geese was increased significantly after infection.Furthermore, significant induction was observed for AvBD6 expression in the Immunohistochemistry.
