为探讨淀粉样前体蛋白( amyloid-β precursor protein, APP)对星形胶质细胞产生的影响,本研究利用APP 基因敲除小鼠(APP KO)和其对应的同窝野生型小鼠(WT),首先通过免疫荧光方法分析了成年小鼠脑内室管膜下区(subventricular zone, SVZ)和皮层区域内的星形胶质细胞(s100β+或者GFAP+/DAPI+)所占的比例。实验数据显示,与WT组相比,APP KO小鼠脑内SVZ和皮层区域内的星形胶质细胞均显著增高;其次,分别分离和培养WT和APP KO小鼠胚胎16.5天SVZ区域的神经前体细胞,诱导分化4天后,统计星形胶质细胞所占比例(GFAP+/DAPI+)和神经前体细胞所占比例(Sox2+/DAPI+),实验结果显示,与WT组比较,APP KO来源的神经前体细胞分化成星形胶质细胞的比例显著升高,并且神经前体细胞的数目显著下降。以上结果表明,在胚胎发育期,APP调控神经前体细胞向星形胶质细胞的分化,从而导致APP KO成年小鼠脑内星形胶质细胞的异常增多。?????

o investigate whether amyloid-β precursor protein (APP) has an effect on astrogenesis, APP knockout mice and their littermates were used in this study. We firstly analyzed the percentage of astrocyte (s100β+or GFAP+/DAPI+) in subventricular zone and cortex in adult APP knockout mice and their controls. Our data indicated that APP knockout (APP KO) mice showed a significant enhancement of astrocytes both in SVZ and cortex. Then, neural progenitor cells (NPCs) from embryonic day 16.5of WT and APP KO mice were isolated and cultured respectively. The ratio of astrocytes to total cells (GFAP+/DAPI+) and the ratio of neural progenitor cells to total cells were measured 4 days after differentiation. Our results showed that more NPCs from APP KO mice differentiate to astrocytes than that from WT mice. And the number of NPCs from APP KO mice was notably decreased compared that from WT mice. Therefore, our data suggested that APP regulates the differentiation of NPCs to astrocytes, thus leading to abnormal increase of astrocytes in adult APP KO mice brain.
