目的 研究SDF-1/CXCR4信号通路在BMP-2诱导小鼠骨髓干细胞迁移中的作用。方法 取对数生长期MSCs细胞,分为SDF-1组、BMP-2组、SDF-1+BMP-2组及BMP-2+AMD3100组(AMD31000浓度均为50μg/L),每组设置0、50、100和200μg/L4个浓度,37℃、5%CO2培养24h,诱导细胞迁移并Transwell检测,倒置显微镜下观察、拍照并用软件Image-ProPlusVersion6.0计数。结果MSCs细胞的迁移与SDF-1和BMP-2密切相关,并且与SDF-1和BMP-2的浓度成正比,当联合使用SDF-1和BMP-2时,细胞迁移数较单独使用时增多,在0μg/L、50μg/L组、100μg/L组、200μg/L组,4种浓度细胞迁移数分别为21.20±2.20,43.90±2.77,72.70±4.79,82.10±4.51,结果比较具有统计学意义(F=553.54,P<0.05),可见当浓度为200μg/L时,MSCs细胞迁移数量最多,并在镜下呈"巢穴"样分布。在当加入50μg/L阻断剂AMD31000时,MSCs细胞迁移数为12.71±1.95,明显低于空白组21.20±2.20,分别加入50μg/L、100μg/L及200μg/L三种不同浓度BMP-2时,细胞迁移数分别为:23.20±2.57,29.50±2.07,40.59±4.25,结果 比较具有统计学意义(F=142.53,P<0.05);结论 SDF-1/CXCR4信号通路为BMP-2诱导MSCs细胞迁移中的重要信号通路之一。?????

Objective To study the role of SDF-1/CXCR4 signaling pathway in BMP-2- induceing the migration of mouse bone marrow stem cell. Methods MSCs in logarithmic growth phase divide into four groups as SDF-1group, BMP-2 group, SDF-1+BMP-2 group and AMD3100 +BMP-2 group, the concentration of AMD31000 were 50μg/L, four different concentrations of 0, 50, 100 and 200μg/L were set, cultured 24h in 37℃, 5% CO2. Cells migration induced and tested under inverted microscope, photograph taken and the number of cells counted with Image-Pro Plus Version 6.0 software. Results The migration of MSCs is closely related to SDF-1 and BMP-2, and proportional to the concentration of both SDF-1 and BMP-2, When SDF-1 and BMP-2 used jointly, the number of MSCs migration was increased significantly. The migration cell number at 0μg/L、50μg/L、100μg/L、200μg/L four different concentration were 21.20±2.20, 43.90±2.77, 72.70±4.79 and 82.10±4.51 respectively, comparison of the results were statistically significant (F=553.54, P<0.05). The number of MSCs cell migration was most when the concentration was 200μg/L, and showed "nests" like distribution In microscopy. when added inhibiter AMD3100, the number of MSCs cell migration was 12.71±1.95, lower than control group 21.20±2.20, when added different concentration of BMP-2, the number of MSCs cell migration were 23.20±2.57, 29.50±2.07, 40.59±4.25 respectively, comparison of the results were statistically significant (F=142.53, P<0.05). Conclusion SDF-1/CXCR4 signaling pathway is an important pathway in BMP-2 inducing the migration of MSCs.
