玉米成熟胚愈伤组织的诱导

试验选取优良玉米自交系K12、昌7-2、137、87-1、478、郑58、综3、178的成熟胚作为外植体,以MS为基本培养基,通过调节培养基中2,4-D浓度和BAP浓度,观察玉米成熟胚愈伤组织的诱导情况,对影响玉米成熟胚愈伤组织的若干因素进行研究,以建立较好的再生体系,为下一步的遗传工程操作奠定基础。实验结果表明,玉米愈伤组织的诱导因基因型的不同有明显的差异,K12诱导率最高达95%,而郑58、综3、178、137诱导率均为0。因此,基因型是影响玉米愈伤组织诱导和培养的一个重要因素。当2,4-D和BAP浓度分别为6mg/L和0.001mg/L时成熟胚愈伤组织诱导率最高达95%,由此可见,玉米成熟胚愈伤组织的诱导率受2,4-D和BAP的影响较大。 关键词:玉米;成熟胚;愈伤组织

Mature dry seeds of eight maize elite inbred lines (K12、478、chang7-2、87-1、zheng58、137、zong3、178、) were used as source of mature embryos (MEs). In order to set up an efficient maize regeneration system ,based on the MS medium ,the author studied the factor that influenced the induction of MEs callus by changing the concentration of maltose and 2,4-D.The result of study indicated that the induction of MEs callus were different from eacher other because of different genotype.The induction of K12 was highest ,and reached to 95% ,While the induction of genotype zheng58、zong3、178、and 137 was zero.Therefore , Genotype was an important factor that influenced the induction and culture of MEs callus.The concentration of maltose and 2,4-D was another factor that influenced the induction and culture of MEs callus.When the concentration of maltose and 2,4-D was 60% and 6mg/L,the induction of MEs callus was height and reached to 95%.
