探讨糖痹康对糖尿病大鼠坐骨神经c-jun及NT-3表达的影响

Objective: To explore the effect of Chinese herbal compound on the expressions of protein and c-jun protein and NT-3 mRNA of diabetic rats' sciatic nerve. Methods: The experimental diabetes mellitus rats model were induced by feeding high fat forage and injection streptozotocin. Tangbikang groups were given Tangbikang with different dose, rats unilaterals sciatic nerve were cutting-up after 16 weeks，immunohistochemical method was used to detect the expressions of NT-3 protein of sciatic nerve, Western blot method was to detect the expression of c-jun protein and the expressions of NT-3 mRNA were detected by semi-quantitative RT-PCR，and compared with mecobalamine group. Results: Compared with normal group, in the model group rats' sciatic nerve, the NT-3 protein and mRNA expression decreased significantly, whereas the c-jun protein expression increased. Tangbikang low, medium and high dose group compared with model group significantly increased the NT-3 protein and mRNA expression (P<0.01), and decreased the c-jun protein(P<0.01). Conclusion: Chinese herbal compound Tangbikang could promote the expressions of NT-3 protein and mRNA, and inhabit the expression of c-jun protein of diabetic peripheral neuropathy rats' sciatic nerve.
