Gh14-3-3L30 affects Verticillium wilt resistance in cotton
黄萎病严重影响棉花品质和产量,棉花黄萎病抗性是由多基因控制的复杂性状,深度挖掘棉花黄萎病抗性关键基因并解析其抗病机制,对于棉花抗病分子育种具有重要意义。课题组前期通过分析接种黄萎病菌的棉花根部转录组数据,筛选到受黄萎病菌诱导表达的Gh14-3-3L30基因,但该基因在棉花黄萎病抗性方面的具体功能和机理尚不清楚。为此,本研究克隆了该基因并构建相关载体进行了功能验证。亚细胞定位分析发现其编码蛋白定位于细胞质,通过病毒诱导的基因沉默(Virus-Induced Gene Silencing, VIGS)技术使棉花中Gh14-3-3L30表达量下降60%,进而进行黄萎病抗性鉴定,与对照组相比,沉默组病情指数显著下降、过氧化氢含量显著下降、POD和木质素的含量显著升高。以上结果表明Gh14-3-3L30通过活性氧和木质素途径负调控棉花抗黄萎病抗性,本研究为棉花抗黄萎病分子育种提供了新的基因资源和理论依据。
Verticillium wilt affects cotton quality and yield seversly. Verticillium wilt resistance is a complex trait mediated by multiple genes. It is of great significance to deeply explore associated key resistant genes and mechanisms for cotton molecular breeding of disease resistance. Preversly, our lab screened Verticillium wilt induced gene Gh14-3-3L30 by analyzing transcriptome data of cotton roots inoculated with Verticillium dahliae. But the resistance function and mechanism of Gh14-3-3L30 were still unclear. For this, we cloned Gh14-3-3L30 and constructed associated vectors to identify its function about Verticillium wilt resistance. Subcellular localization showed that Gh14-3-3L30 localized in the cytoplasm. The expression of Gh14-3-3L30 was silenced to 60% by virus-induced gene silencing (VIGS) in cotton and then the Verticillium wilt resistance was identifed. Compared with the control group, the disease index of the silenceD group was significantly decreased, and the contents of hydrogen peroxide, POD and lignin were significantly increased. Our results suggested that Gh14-3-3L30 might be involve in the process of Verticillium wilt resistance by regulating the content of ROS and lignin, which provided new genetic resources and theoretical basis for molecular breeding of Verticillium wilt resistance in cotton.
