目的 探讨急性髓系白血病(AML)细胞来源的树突状细胞(DC)上CD274基因的表达水平及影响其表达的因素。方法 应用流式细胞仪及实时定量PCR法(RQ-PCR)检测AML患者发病时AML细胞来源DC(AML-DC)及缓解后骨髓单个核细胞(MNC)来源DC(MNC-DC)上CD274的蛋白及mRNA表达水平。并比较不同细胞因子对其表达水平及功能的影响。结果 1、AML-DC上CD274呈高水平表达,诱导缓解后MNC-DC上CD274表达水平显著下降。2、γ-干扰素(IFN-γ)、白细胞介素(IL)-10等细胞因子可以增高CD274的表达水平。3、中和性抗体封闭CD274的表达后AML-DC刺激异基因T细胞的增殖能力提高、IL-12分泌量增加。结论 AML-DC上高水平表达CD274,通过阻断CD274的表达有助于提高AML-DC的功能。

Objective To Analysis the Expression level and Influencing Factors of CD274 on dendritic cells drived from acute meyloid cells.Methods the CD274 mRNA and molecular expression levels of acute leukemia cell derived dendritic cells from newly diagnosed leukemia patients were detected by rea1time quantitative PCR and flow cytometry.Results 1.AML cell can be induced into dendritic cells with high level of CD274 mRNA and molecular. CD274 on DC derived from bone marrow mononuclear cell of patients achieved CR was significantly lower than that on AML-DC.2. IFN-γ、IL-10 significant increase the CD274 on AML-DC. 3.After the CD274 was blocked with antibody, the allostimulatory abilities of AML-DC were significantly increased.Conclusion The immunological activity of AML-DC can be increased through blocking the CD274.
