Inhibitory effect of Ethanol Extract of Hedyotis diffusa Willd on colorectal cancer drug-resistant xenograft in nude mice

探讨白花蛇舌草乙醇提取物(EEHDW)对大肠癌耐药裸鼠移植瘤生长的抑制作用及其分子机制。通过构建大肠癌5-氟尿嘧啶(5-FU)耐药细胞皮下移植瘤动物模型,随机分为对照组、EEHDW组、5-FU组、EEHDW联合5-FU组(联合组),通过瘤体体积测量、RT-PCR和IHC等方法计算抑瘤率、检测移植瘤组织中PCNA的表达及CyclinD1、CDK4、p21的mRNA和蛋白表达。与对照组比较,5-FU对移植瘤的生长无明显影响(P>0.05),EEHDW及联合组移植瘤生长显著抑制(P<0.05),且联合组抑瘤效果更显著;EEHDW及联合组明显抑制移植瘤细胞增殖,下调移植瘤组织中Cyclin D1、CDK4、上调p21的mRNA和蛋白表达(P<0.05)。 EEHDW在体内通过抑制Cyclin D1、CDK4、上调p21表达是其抑制大肠癌耐药移植瘤生长的重要机制。

nude mice drug-resistant subcutaneous xenograft model was established to study the growth inhibitory effect of Ethanol Extract of Hedyotis diffusa Willd (EEHDW). Mice were randomized into 4 groups (Control, EEHDW, 5-FU, and EEHDW-5-FU combination), to calculate the inhibition rate by determination of tumor volume, and to detect PCNA by IHC, and CyclinD1, CDK4 as well as p21 by RT-PCR and IHC. Compared with the control group, 5-FU had few effect on tumor growth (P>0.05), while EEHDW and combination treatment significantly inhibited growth of xenograft (P<0.05), and combination treatment had a better effect; EEHDW and combination treatment significantly inhibited the proliferation of tumor cells, down-regulated cyclinD1, CDK4, and promoted expression of p21 on mRNA and protein levels (P<0.05). The down-regulation of cyclinD1, CDK4 and up-regulation of p21 are important mechanisms by which EEHDW inhibits colorectal cancer drug-resistant xenograft.
