外源Wolbachia人工转染后的滴度变化可能影响其控制新宿主种群的能力。首先将来自黑腹果蝇的wMel菌株通过显微注射的方式人工水平转移到烟粉虱中;然后采用荧光原位杂交证实wMel在新宿主中的存在;再建立携带wMel的烟粉虱单雌系;最后采用实时荧光定量PCR检测wMel在烟粉虱后代中的滴度变化。结果表明,wMel菌株的滴度在转染后第2-3代达到一个低点后缓慢上升,至第6代达到峰值,然后下降,至第9代达最低点,然后出现小幅反弹。该结果提示,wMel菌株可在烟粉虱种群中传播,但其滴度在各代中波动较大,这似乎影响了wMel菌株在转染后代种群中的传播。
he change in the titre of a foreign Wolbachia strain after transinfecton may be involved in its capability for controlling host population. Herein, the wMel Wolbachia strain from the fruit fly Drosophila melanogaster was artificially transferred into the whitefly Bemisia tabaci. Fluorescence in situ hybridization verified the presence of wMel in the new host; the transinfected whiteflies were then used to establish the isofemale lines. The results based on real-time quantitative PCR showed that the titre of wMel strain reached a low level at G2 and G3 after transinfecton, then rose steadily and reached the peak at G6, but dropped to the minimum at G9 and then slightly recovered after G10. These results suggest that wMel can be transmitted in B. tabaci populations, but its titre fluctuated among different generations, which seems to have affected the transmission of wMel in transinfected offspring populations of B. tabaci.
