Effects of Addition of Green Tea Polyphenols on Early Development Bovine Embryos during In Vitro Fertilization and In Vitro Culture

为了研究绿茶多酚(GTPs)对奶牛卵母细胞体外受精和植前胚胎早期发育的影响,分别在卵母细胞体外受精(IVF)和IVF胚胎培养期间向培养液中加入不同浓度的(0、10、15、20、25 μM) GTPs。结果表明:在IVF培养液中添加不同浓度的GTPs均不能提高卵裂率及囊胚率;添加15μM GTPs到胚胎培养液中显著提高了囊胚率,但添加更高浓度的GTPs(20 或 25 μM)并没有进一步增加囊胚率;与对照组相比,添加15μM GTPs到胚胎培养液中显著提高了第7d囊胚内的GSH含量和GR基因的表达水平,但添加更高浓度(25μM)GTPs反而降低了囊胚内GSH含量和GR基因的表达。结果提示,在胚胎体外培养液中添加适量浓度GTPs可缓解氧化应激对体外胚胎的损伤,显著提高胚胎早期发育能力。

he objective of this study is to examine the effects of green tea polyphenols (GTPs) on in vitro fertilization (IVF) and early embryo development in cow. The different concentrations of GTPs (0, 10, 15, 20 and 25 μM) were added respectively into media during IVF and in vitro culture (IVC) of embryos. The results show that the cleavage rate and blastocyst rate were not significant difference when GTPs are added in IVF media. Addition of 15 μM GTPs to IVC media significantly increased the blastocyst rate, but no further increase in the blastocyst rate when higher concentrations GTPs (20 or 25 μM) are added. Compared with the control group, addition of 15μM GTPs to IVC media significantly increased the GSH content and GR gene expression levels within 7d blastocyst, but supplementation of a higher concentration (25μM) GTPs reduced the GSH content and GR gene expression. These results suggest that addition of appropriate concentration of GTPs can improve the ability of early embryonic development by alleviating oxidative stress during IVC period.
