为了研究蛇含委陵菜(Potentilla kleiniana)的化学成分及其抗炎活性,该文利用 D-101 大孔树脂、硅胶及 Toyopearl HW-40F 等色谱技术对蛇含委陵菜 60%乙醇提取物进行分离纯 化,通过核磁共振波谱(NMR)、高分辨质谱(HR-ESI-MS)鉴定化合物的结构,采用小 鼠巨噬细胞(RAW 264.7)体外炎症模型评价化合物的抗炎活性。结果表明:(1)从蛇含 委陵菜中分离得到 15 个化合物,分别鉴定为 2-(heptadecanoyloxy)propane-1,3-diyl distearate (1)、9,12,13-三羟基-10,15-十八二烯酸(2)、9,12,13-三羟基-10,15-十八二烯酸甲酯 (3)、2,2'-氧代双(1,4-二叔丁苯)(4)、大黄素(5)、大黄酚(6)、(6R,9R)-3-酮-α- 紫罗兰醇-9-O-β-D-吡喃葡萄糖苷(7)、新穿心莲内酯(8)、甲基-α-D-呋喃果糖苷 (9)、1-O-β-D-吡喃果糖-α-D-吡喃阿洛糖苷(10)、对香豆酸(11)、cesternosides A (12)、koaburaside(13)、荭草素(14)、异荭草素(15),均首次从委陵菜属植物中分 离得到。(2)抗炎实验结果显示,化合物 1-3、8、11-15 具有一定的 NO 释放抑制活性, 其中化合物 8 在 25 μmol·L-1 浓度下抑制率为72.5%。该研究丰富了蛇含委陵菜的植物化学 信息,明确了脂肪酸衍生物、酚性成分及二萜类成分是其抗炎活性成分,为蛇含委陵菜的 进一步开发利用提供理论依据。

he purpose of this paper is to investigate the chemical constituents of Potentilla kleiniana and their anti-inflammatory activity. The 60% ethanol extract of P. kleiniana were isolated by D-101 macroporous adsorptive resins, silica gel, Toyopearl HW-40F and other methods, and their chemical structures were elucidated on the basis of NMR and HR-ESI-MS analysis. Meanwhile, the anti-inflammatory activity of the compounds was evaluated by mouse macrophage (RAW 264.7) inflammatory model induced by lipopolysaccharide (LPS) in vitro. The results were as follows: (1) Fifteen compounds were isolated and identified from P. kleiniana as 2- (heptadecanoyloxy)propane-1,3-diyl distearate (1), 9,12,13-trihydroxy-10,15-octadecadienoic acid (2), methyl-9,12,13-trihydroxyoctadeca-10,15-dienoate (3), 2,2'-oxybis(1,4-di-tert-butylbenzene) (4), emodin (5), chrysophanol (6), (6R,9R)-3-oxo-α-ionol-9-O-β-D-glucopyranoside (7), neoandrographolide (8), methyl-α-D-fructofuranosides (9), 1-O-β-D-fructofuranosyl-α-D-allopyranos (10), p-coumaric acid (11), cesternosides A (12), koaburaside (13), orientin (14), isoorientin (15). Compounds 1-15 were obtained from Potentilla genus for the first time. (2) The anti-inflammatory test results showed that compounds 1-3, 8, and 11-15 had moderate inhibitory activities on NO production, and the inhibition rate of 8 was 72.5% at the concentrations of 25 μmol·L-1. In conclusion, the study enriches the phytochemical information of P. kleiniana, and clarifies that fatty acid derivatives, phenolic components and diterpenoids are anti-inflammatory active components, which provide a theoretical basis for further exploitation of P. kleiniana.
