HPLC测定黄柏不同部位盐酸小檗碱和盐酸药根碱
etermination of berberine hydrochloride and jatrorrhizine hydrochlo-ride in different parts of Cortex Phellodendri Chinensis by HPLC
目的：采用HPLC法同时测定黄柏枝皮、干皮、根皮中药根碱、小檗碱的含量。方法：通过考察不同溶剂、不同提取方法、不同提取时间和不同流动相，以药根碱、小檗碱提取率为指标，采用高效液相色谱法进行测定。结果：以盐酸-甲醇（1：100）超声提取30min，采用流动相0.05 mol／L磷酸二氢钾缓冲液(磷酸调pH值至3.0)一乙腈(72:28)，进样量20μL，检测波长265 nm，室温条件下检测。结论：黄柏枝皮、干皮、根皮中生物碱含量有明显差异。
Objective The contents of berberine hydrochloride and jatrorrhizine hydrochloride in cortex of branch、trunk and root of Phellodendron chinense Schneid. were measured by HPLC.Methods The content of berberine and jatrorrhizine was assayed as examination marker by HPLC,different extracting solvents、methods and time, and different mobile phase was investigated.Results The sample was treated by ultrasonic extraction with HCL一MeOH solution(1:l 00,by volume)．The filtrate was separated by using acetonitrile—0.05 mol／L KH2 PO4(28：72，by volume)solution，whose pH was adjusted to 3.0 with H3PO4，as mobile phase，and detected at 265 nm，25℃.Conclusions The contents of alkaloids in cortex of branch、trunk and root were different remarkably.
