刺参DLC2基因克隆及其夏眠期间表达特征的研究

动力蛋白轻链是动力蛋白的组成部分,在细胞质内大分子物质的转运中发挥重要作用。本研究利用通过RACE、GENOME WALKING等技术测定了刺参Apostichopus japonicus (Selenka) 动力蛋白轻链dynein light chain 2, cytoplasmic isoform X2 (DLC2)基因结构和在夏眠不同阶段表达量变化,为进一步的刺参夏眠分子调控机理研究奠定基础; 研究结果表明刺参DLC2基因cDNA全长804bp,编码89个氨基酸,并预测得到具有一个Dynein light保守结构域;刺参DLC2与其他物种基因同源性和进化树分析显示该基因具有高度保守性。刺参DLC2基因含有2个外显子和330bp的启动子区域,并预测到多个转录因子结合位点。qRT-PCR研究表明,刺参肠道和肌肉中,DLC2基因在深度夏眠阶段表达量分别表现出显著上调和下调,而在呼吸树中表达量变化不显著。
ynein light chain is part of dynein which plays an important role in the transport of macromolecules in the cytoplasm. The present study amis to explore the gene structure and expression character of dynein light chain 2, cytoplasmic isoform X2 (DLC2) in sea cucumber Apostichopus japonicus (Selenka) during aestivation using RACE, genome walking and qRT-PCR technologies. The deduced amino acid sequences and characteristic motifs of sea cucumber DLC2 showed high homology to those of their mammalian counterparts. DLC2 gene contains 2 exons and a 330bp promoter, and several transcription factor binding sites were predicted. qRT-PCR analysis indicated that the expression levels of DLC2 were significantly up-regulated in intestine and significantly down-regulated in muscle during aestivation. .
