氟虫腈对斑马鱼miR-155及部分预测靶基因表达影响
Effects of fipronil on zebrafish miR-155 and some predicted target genes
近年来以生物化学与分子生物学方法来研究农药的作用机制正逐渐增加,研究表明生物分子如DNA,RNA等会受农药的影响。MicroRNAs(miRNAs)是一类在基因表达中起重要调控作用的长约22nt的非编码小RNA分子。研究表明氟虫腈能影响斑马鱼miR-155的表达,而对其使相应的靶基因表达发生改变的可能性尚不明确。本研究通过数据库筛选出斑马鱼miR-155的3条可能靶基因(cyb561d2、LOC570587和zgc158824),用荧光定量PCR的方法检测它们相对于miR-155的表达变化情况,结果显示,在氟虫腈处理组miR-155表达下降的情况下,斑马鱼眼中基因cyb561d2表达量大幅上升,约6倍于空白组, LOC570587表达略有上升,约2倍,变化不明显, zgc158824表达基本无变化。在斑马鱼脑中cyb561d2表达上升,约为空白组4倍,LOC570587表达下降,为空白组的0.8倍,zgc158824表达仍无变化。
Recent years,many research on mechanism of pesticides has been growing through biochemistry and molecule biology methods. Report shows that biomolecules such as DNA and RNA can be effected by pesticides.MicroRNAs(miRNAs) are small RNAs with the length of about 22nt,which play an important role in regulation of gene expression.Data shows that expression of zebrafish miR-155 could be changed by fipronil,while its effect on posible target genes is still unknown.Three potential target genes of zebrafish miR-155(cyb561d2、LOC570587 and zgc158824 )was chosen from databases, Expression of these genes relative to miR-155 was tested by real-time PCR. Result shows,when expression of zebrafish miR-155 in zebrafish eyes was downregulated in the fipronil group,gene cyb561d2 was upregulated obviously-about six times, LOC570587 upregulated about two times, zgc158824 showed hardly any change.While in their brains, cyb561d2 was upregulated about four times, LOC570587 was downregulated about 0.8 times, zgc158824 also had little change.
