重吸附法回收利用纤维素酶的工艺研究
Optimization of cellulase re-adsorption for reutilization
以玉米芯为底物,分别考察了Tween 80浓度、加酶量和底物浓度对纤维素酶回收利用的影响规律,结果表明,酶回收率随Tween 80浓度和加酶量的增加呈先增大再减小的趋势。此外,酶回收率随底物浓度的增加而逐渐增加。确定重吸附法回收纤维素酶的最优工艺条件为:Tween 80浓度:0.25 g/g 纤维素;加酶量:40 FPU/g 纤维素;底物浓度:3.6 %. 在最优条件下,酶回收率可达83.0 %。该研究为降低纤维素酶成本比例提供了有效的解决方法。
he effect of Tween 80 addition, enzyme loading and substrate concentration on the re-adsorption of cellulases, which hydrolyzed the corncob after the pretreatment by soaking in aqueous ammonia, was studied in this paper. As the addition of Tween 80 and enzyme loading increased, the cellulase recovery first increased and then decreased gradually. The cellulase recovery increased gradually with the increasing concentration of the substrate. The optimum conditions were drawn as follows: Tween 80 addition of 0.25 g/g cellulose, enzyme loading of 40 FPU/g glucan, substrate concentration of 3.6 wt%. Under this condition, 83.0 % cellulase was recovered. This research provided a good solution to reduce the cellulase cost.
