慢性高眼压大鼠动物模型的视网膜和视网膜神经节细胞变化的研究
Study of retina and retina ganglion cells in a rat model of chronic ocular hypertension
目的 观察高眼压对大鼠视网膜和视网膜神经节细胞的损伤。方法 将60只大鼠随机分为手术组和空白对照组。采用烙闭巩膜上静脉的方法建立慢性高眼压模型,用Goldmann鼠眼压计在造模前测定基础眼压。造模后测量各组大鼠第l、3、5、7、14、28和60天的眼压,检测大鼠视网膜神经节细胞数量,测量视网膜内丛状层的厚度。结果 烙闭3条巩膜上静脉的方法能成功建立慢性高眼压模型。高眼压状态下,大鼠视网膜神经节细胞的数量明显变少,并且发生空泡样变,视网膜内丛状层的厚度也明显变薄。结论 慢性高眼压状态下,大鼠的视网膜变薄,视神经节细胞明显变少并凋亡。
Purpose To investigate the effect of IOP on retina and retina ganglion cells in a rat model of chronic ocular hypertension.Methods Chronic ocular hypertension was induced by three episcleral vein cauterizations (EVC) in rats. IOP was measured one day before operation and at 1, 3, 5, 7, 14, 28, 56 days after operation. Retinal histologic sections were measured to determine inner plexiform layer (IPL) thickness and the number of cell bodies in the ganglion cell layer (GCL). Results Cauterizations caused IOP increase over 73% that was maintained for 2 months. High IOP decreased the number of retina ganglion cells and the thickness of IPL. Conclusions High IOP can make retina ganglion cells loss and decrease the retinal thickness in a rat model of chronic ocular hypertension.
