肝细胞生长因子对人肝癌细胞增殖作用及机制

Objective To investigate the effect of hepatocyte growth factor （HGF）on the proliferation in human hepatocellualr carcinoma cells. Methods Human hepatocellualr carcinoma cells HepG2 were treated with different concentrations of HGF for different times, and the proliferation of these cells were examined by colorimetric BrdU cell proliferation enzyme-linked immunosorbent assay. The expression of cyclin dependent kinases inhibitors, p27 and p18, was examined . Results HGF inhibited the proliferation of hepatoma cells in a dose and time dependent manner. The expression of p27, but not p18, was significantly induced by HGF. Conclusion This study suggested that HGF could inhibit HepG2 cell proliferation, and the up-regulation of p27 might account for this suppressed proliferation.
