目的:探讨健脾益气摄血方对ITP模型小鼠血清β-内啡肽(β-EP)、血管活性肠肽(VIP)、5-羟色胺(5-HT)、去甲肾上腺素(NE)、缩胆囊素(CCK)的影响。方法:以免疫造模法建立ITP小鼠模型,以强的松、健脾益气摄血方为干预药物,用酶联免疫吸附法(ELISA)检测模型小鼠血清β-EP、VIP、5-HT、NE、CCK含量变化。结果:①β-EP:与空白对照组比较,模型组小鼠血清β-EP含量升高,具有统计学意义(p<0.05);与模型组比较,强的松干预后小鼠血清β-EP含量变化无统计学意义(p>0.05),健脾益气摄血方干预后,有统计学意义(p<0.05)。②VIP:与空白对照组比较,模型组VIP含量有明显降低,有统计学意义(p<0.05);与模型组对照,强的松组、健脾组VIP含量明显回升,有统计学意义(p<0.05)。③5-HT:与空白组对照,模型组5-HT含量有显著降低,有统计学意义(p<0.05);与模型组比较,强的松组(p<0.01)、健脾组(p<0.05)含量显著回升。④NE:与空白对照组比较,模型组、强的松、健脾组小鼠血清NE含量变化有统计学意义(p<0.05)。⑤CCK:与空白对照组比较,模型组、强的松、健脾摄血组小鼠CCK含量变化均无统计学意义(p>0.05)。结论:β-EP、VIP、5-HT、NE等神经递质或血管活性物质参与了ITP小鼠模型发病过程,益气健脾摄血方对ITP模型小鼠血清β-EP、VIP、5-HT、NE含量有明显的调控效应,而CCK未显示参与ITP模型发生过程。

Objectives: To investigate the effect of prescription JIANPIYIQISHEXUE on β-EP, VIP, 5-HT, NE and CCK in serum from ITP mice model. Methods: The ITP mice immune model was established. Prednisone and prescription JIANPIYIQISHEXUE (granules) were given, and the changes of β-EP, VIP, 5-HT, NE and CCK in serum were detected using ELISA method. Results: ①β-EP: β-EP in serum was up-regulated in ITP group (p<0.05); compared with ITP group, there was no significant change in prednisone group (p>0.05), but was recovered in JIANPIYIQISHEXUE group (p<0.05). ②VIP: VIP in serum was down-regulated in ITP group (p<0.05); but was recovered both in prednisone group and JIANPIYIQISHEXUE group (p<0.05). ③5-HT: 5-HT from serum was reduced in ITP group (p<0.05); and were augmented in prednisone group (p<0.01) and JIANPIYIQISHEXUE group (p<0.05). ④NE: The content of NE increased in ITP group and also after administration (p<0.05). ⑤CCK: The content of CCK in serum had no significant change after modeling and administration. Conclusions: The neurotransmitter and vasoactive substances including β-EP, VIP, 5-HT, NE were involved in the pathogenesis of ITP mice model, prescription JIANPIYIQISHEXUE effectively regulate the content of β-EP, VIP, 5-HT and NE. CCK did not participate in the process of ITP model.
