六味地黄汤对快速衰小鼠认知功能及Wnt3a、GSK-3β表达的影响

目的:研究六味地黄汤对D-半乳糖(D-gal)致快速衰老小鼠认知功能及Wnt3a、GS K-3β表达的影响。方法:以500mg/(kgod)的D半乳糖颈部皮下注射复制快速衰老小鼠模型,将动物随机分为正常组、模型组、六味高剂量组、六味中剂量组、六味低剂量组、VitE组,给予不同的处理。灌胃体积为(0.1ml/ 10g),历时8周,观察小鼠的体质量变化,行Morris水迷宫试验评价小鼠的认知功能,免疫组化法、RT-qPCR法检测Wnt3a、GSK-3β的表达。结果:与正常组及其他组比较,模型组的体质量增重少(P<0.05),潜伏期长和垮台次数多(P<0.05);六味地黄汤高剂量组与正常组比较,体质量增重、潜伏期以及垮台次数差异(P>0.05)无统计学意义;六味地黄汤低剂量组与VitE组比较体质量及学习记忆能力亦无统计学意义(P>0.05);六味高组与其他用药组比较具有统计学意义(P<0.05);免疫组化法与RT-qPCR的检测结果有高度一致性,模型组Wnt3a的表达少,GSK-3β活性高,与正常组比较差异有统计学意义(P<0.01),高剂量组Wnt3a高表达,GSK-3β活性低与中低剂量组、VitE组比较有高度统计学意义(P<0.01)。结论:六味地黄汤可以有效改善D-半乳糖致快速衰老小鼠的学习记忆障碍;增加衰老小鼠Wnt3a的表达,激活Wnt信号通路,下调GSK-3β的表达可能是六味地黄汤延缓衰老健脑益智的机制。?????
Objective:To study the effects of Liuwei dihuang decoction on recognition in aging mice of D - galactose (D - gal) and the express of Wnt3a、GSK-3β.Methods: The rapid aging animal models was established by d-galactose subcutaneously injected drugs as 500 mg/(kg o d), the animals were randomly divided into normal group, model group, the high , Middle and low dose group of Liuwei dihuang decoction, VitE group (positive control group), to give different treatment. Lavage for volume (0.1 ml / 10 g), eight weeks, to observe the mice body quality change, line Morris water maze test evaluation of cognitive function in mice,The expression of Wnt3a and GSK-3βwas detected by immunohistochemical method and RT-PCR method. Results: Compared with the other groups, model group's quality of body weight (P < 0.05), less down longer latency and more (P < 0.05); Liuwei dihuang decoction group compared with normal group, there was no significant difference betweenthe high quality of body weight, the incubation period, and the collapse of frequency (P > 0.05);Low dose group of Liuwei dihuang decoction compared with VitE group of body quality and the ability of learning and memory also have no statistical significance (P > 0.05);The results were highly consistent with the detection of RT-qPCR, In model group, the expression of Wnt3a less, GSK-3βactivity,compared with normal group and treatment group difference was statistically significant (P < 0.01), High expression of Wnt3a in high dose group, GSK-3βactivity was lower than low dose group and VitE group, and it was highly significant (P<0.01)., Conclusion:liuwei dihuang decoction can improve the D - galactose rapid aging mice learning memory disorders; Increased expression of Wnt3a in aging mice, activation of the Wnt signaling pathway, down GSK-3beta expression may be the Liuwei Dihuang Decoction delaying senescence of Jiannao Yizhi mechanism.?????
